VOCAL EXTRACTION
It's best to have both a main track and an accompanying instrumental track. If so, you can use Utagoe (http://www.vector.co.jp/downl oad/file/win95/art/ff258894.ht ml). I've had best results with the 1.1 setting in general. This sometimes does not work for various reasons.
If there's no instrumental, it's much harder. You can extract the center channel of a single track (see below), then use an equalizer to suppress non-vocal frequencies.
INSTRUMENTAL EXTRACTION
To extract an instrumental track where one does not exist, you can use the dsp_centercut plugin (http://moitah.net) for Winamp. It has several settings. Choose the Sides (Bass to Sides) setting for best results. Some tracks may be harder to work with than others.

I've used the following audio editing applications: GarageBand, Audacity, FL Studio, Melodyne.
