preparing four different media slides with a water sample taken from my Koi pond.
to produce a total bacteria count

The healthy value of total bacteria count in a koi pond at any given time should not exceed 5000 cfu and below 1000 cfu is considered a very good count; the maximum allowed total count value for tap water here in holland for instance is 100 cfu.

A healthy value or low total count value wil not leed to any kind of bacterial problem, in case a koi hurts himself and has a wound you wil witnes it healing without any problems of inflamation within a few days.
Where with a high total count there often is a inflamation of the wound meaning it is infected by bacteria.

You could conclude that using a swab and making a total bacteria count is a useful preventive measure even if you can't determine the difference between good and bad bacteria.

In this case we can differentiate some bad species with the plates we use being: # Coliform bacteria # E. coli # Enterobacteria # Listeria # Salmonella # Staphylococcus aureus # Total aerobic count

And some more being developed but in al cases the total count is a very good indication of the bacterial Health status in your pond, even with total counts below 25 you will have a fully functional (Bacterial) filter system.
