Malaria 96 well high-throughput Luciferase assay. Author: Flor Caro DeRisi Lab UCSF Malaria parasites transfected with a renilla luciferase reporter construct are released from the red blood cells (RBCs) by saponin lysis. Excess hemoglobin is washed by vacuum filtration on a 96 wel PVDF filter plate while parasites are retained. Parasites are lysed using the renilla luciferase lysis buffer and collected by centrifugation on a white assay plate. RLUs are measured on a luminometer.
