On the opponent's first turn, they'll likely lay down two or even three magic/traps. I've noticed that the traps that people lay down first are always the most powerful, because there is no doubt in their mind that it will work for them in the short term. The ones they lay second or third tend to be more situational, but may benefit them later. I've also noticed that when a person draws a card and instantly sets it face down in the trap/magic slot without needing to read it over or think about it, it's usually a very simple but powerful trap, like magic cylinder, trap hole, mirror force, etc. If it's something situational, they'll spend more time looking at their cards, and their other cards. And if they're looking at the graveyard, then it you can be sure they're thinking of bringing something back from the grave. So basically if you have MST or any other cards that let you destroy spell/traps, if they're face down, go for the cards that they set down the quickest. Generally if they slapped it down quicktime and end their turn, then that trap is the most deadly. Just a few patterns I've noticed!
