This tutorial shows how to use Bio-Rad's PDQuest 2-D analysis software with the Bio-Rad EXQuest spot cutter to automatically excise protein spots from 2-D electrophoresis gels and place the gel plugs into the wells of a microtiter plate for enzymatic digestion and mass spectroscopy analysis.
The narration covers how to select the analysis set for excision, the gel(s) to be cut, the gel type, the size of the cutting tip, and the number of cuts to make in order to maximize protein extraction. The video also covers how to select the specifications of the microtiter plate and what wells to fill with the plugs, as well as how to export a list of cut spots and relative quantities. See how the gel can be exactly positioned in the spot cutter by comparing the patterns of the original gel image with the actual gel inside the spot cutter. Color-coded spot matching enables visual confirmation of precise alignment.
