Im sure my Lexus IS300 brake disc is not meant to do this?! Think it is missing a screw?!

Lol, that's normal. Once wheel is installed it'll keep the rotor in place.

...that's fine.  when the lugs (which are conical at one end) bite down, it centers the rotor.  as for it moving in and out, that's from the pistons not being engaged fully (which is what you want).  they will close the first time you use them. (master toyota tech here)

its fine once you put the wheel on its not going to move.
