This video tutorial demonstrates the loading and operation of Bio-Rad's MicroRotofor cell for a focusing run, starting with a previously assembled focusing chamber and electrodes. Following isoelectric focusing, Bio-Rad's colored protein standards provide a visual reference for the degree of separation. The MicroRotofor cell offers rapid, simple, and effective fractionation and enrichment of proteins from complex protein mixtures.
Due to the great complexity of samples such as tissues, cells, and body fluids, protein researchers may want to fractionate crude samples prior to analysis by two-dimensional (2-D) gel electrophoresis. Fractionation is particularly relevant when trying to detect low-abundance proteins in samples like serum, cerebrospinal fluid (CSF), and urine because of the presence of highly abundant proteins such as albumin and immunoglobulins. The MicroRotofor cell uses liquid-phase IEF to fractionate small volumes (2.5 ml) of proteins in free solution based on isoelectric point (pI). Protein fractionation by pI improves sample loading and separation on narrow- and micro-range IPG strips in downstream 2-D electrophoresis applications.
