【一般】ある神経細胞集団を黄色蛍光タンパク質(YFP)の蛍光で標識した形質変換マウスをホルマリン固定し、その全脳を作動距離4ミリメートルの対物レンズを備えた二光­子励起蛍光顕微鏡で観察。脳表面から大脳皮質、白質、海馬CA1、歯状回へと奥深くまで観察することに成功。

Visualizing the 3D architecture of neuronal networks comprised of proteins expressing yellow fluorescent protein (YFP) in a very long quadratic prism (4 mm). A series of X-Y images through the 3D reconstruction data (500 × 500 × 4,000 μm volume) from the cerebral surface to the dentate gyrus of the YFP-H mouse (13 weeks old). TPEFM with a non-descanned detector and a custom designed 25× objective lens (NA 1.0, WD 4.0 mm) was used.
