If you are comparing the first mans shot and Derrens, the recoil is about the same, so your theory is no-go. And the bullet fits the gun if you wasn't borned yesterday :)

fake gun, he had no recoil when he shot that. The bullet size was big too. I've shot real guns before and if the caliber is that big, hehe ur hand will jump 10in up wards with a single hand at least.

Aside from his suggestions, I think he was picking up his micro-expressions to make sure. He told him to keep repeating the number to himself.

If you listen carefully to the things Derren says from 3:15 till he starts the game, it's obvious that Derren is trying to make the person choose chamber number one. He's invading his unconciousness with sentences like "I want you to choose ONE of those numbers", "Look at them now and CHOOSE ONE" and "Are you thinking of ONE now?" He's probably been doing this from the moment they met eachother. ;)
