Creating in CMYK is fine, I like to create in RGB and convert to CMYK to see which colors are affected the most. Most colors don't change much when converted to CMYK.

I use an Epson PhotoStylus 2200 large format printer. You don't need to use an Epson, there are other printers that work just as good. But Epson in my option is the best, there is UV protectors in the ink that just make the transparencies work alot better than other printers would. When i started out I just took my PDF's to Kinkos and had a few of the same transparencies printed of each design, then just tapes a few together. Hope it helps!

I put up a tutorial for CMYK separations using Photoshop on my website. The address for my site is in the video description. Let me know if this helps.

@XsplisiT You will probably not get great results using a 110 mesh screen. I print my CMYK using 305 mesh, but a high 200 mesh would also work.
Divide your mesh count from something in-between 3.5 and 5 to get your line angles. The frequency takes some playing around with . You might have luck using one angle for all, or staggering the angles by 30 degrees.
Good Luck.
