Piwi protein is required to silence transposons in both the germline and somatic cells of the gonad. The RNA helicase Armitage, which is expressed in the germline, is required for Piwi localization and function in germline cells only. In Armitage mutant flies, Piwi-associated piRNAs corresponding to transposons regulated in the germline were lost. The movie shows Z-stacks of a Drosophila Armitage mutant egg chamber stained for Piwi (green), Lamin (red, which marks the nuclear periphery) and DNA (blue, which marks the nucleus). Notably, Piwi staining is absent from the central, germline nuclei, compared to its intense staining in the surrounding, follicle cell nuclei.
