SeCore® DRB1 Locus Exon 2 & 3 Sequencing HLA Kit -- Watch this short video to see the significant difference this new DRB1 kit could make for your SBT DRB1 analysis.

Life Technologies is committed to simplifying the complexity in HLA typing analysis, and the new SeCore® DRB1 Locus Exons 2 & 3 kit does just that.

This new kit is the first and only sequence-based DRB1 HLA typing kit to offer complete bi-directional sequencing of DRB1 exon 2 and 3.

Using uTYPE® SBT HLA analysis software let's review a typical DRB1 sample analysis when exon 2 coverage is limited compared to when full exon 2 coverage is provided.

When starting the analysis process, notice the green exon 2 bar at the top. The gap at the beginning and at the end tells us full coverage is not provided. Let's review the impact of this limitation on our analysis.

Within the match window list, on the left we can review the number of mismatches for this sample. As we scroll down, we notice a significant number of ambiguities. In this case, there are over 75.

Now, let's change the settings in the software to see the difference full exon 2 coverage makes for sample analysis in this case. To change the settings, click the preferences icon at the top of the screen.

Click the "Exon Trim" option

Change the "default" setting to "full exon" in the option list for both exons 2 and 3.

Click "apply changes".

Then click "exit" to leave the preferences.

With full exon 2 coverage, the number of ambiguities within the match list has decreased from over 75 to 4.

Let's take a closer look at the quality of data for the first hypervariable region of exon 2 since this is the area of the DRB1 gene that is most polymorphic and the area that causes most of the ambiguous results.

The new SeCore DRB1 kit is designed to provide clean and clear bi-directional sequencing data in the first hypervariable region.
And now across the entire exon 2.

When reviewing the sequencing data for exon 3, we see there is complete bi-directional coverage with clean and clear sequencing data.

The new SeCore® DRB1 kit includes a GSSP for Codon 86, so let's review this data Click "GTG" to review this data.

The Codon 86 GSSP data shows 8 alleles.

Now, we will use the "filter" function, to narrow the results in uTYPE® software even further. The analysis now shows only 2 results.

This sample analysis shows the advantages of the new SeCore® DRB1 exon 2 &3 kit because it offers complete bi-directional sequencing which dramatically reduces the number of ambiguities. And, greater first pass resolution means less time and expense is spent on secondary verification methods and final typing results are generated with sequencing data in less time.
