It is best to let them dry for at lest 24 hours. They will cure after 10 days but you can use them before that. No sealer required. If you did it woudl dull the beads.

These glasses are unique and a party pleaser. Do NOT put them in the dishwasher! I have some glasses I made 5 tears ago. Becasue we actually use them beads do fall off occasionally. I suggest each tiem before you use them just rub your hand over them and any loose beads will come off.
