I think the reason Nat freaked out about Mrs. Jennings and everyone thinking they're a couple is because she was so affected by that kiss. I think she would have laughed and brushed it off otherwise.

That kiss slammed it home for these two! I think that up to this point they had feelings for each other but just didn't realize it till now. It is more vivid now and more obvious for them on how they feel.
