No, if your stud is behind the hole then you can still use a butterfly patch in it ,but I would advise you to use a couple of drywall screws to secure the patch to the stud for extra stability. You do not have to do this, but you can't go wrong securing it. If you have a large hole between two studs,I would advise you to cut your piece to fit stud to stud or use blocking behind the hole.
