2D-PAGE is the most popular protein separation technique which excels in terms of resolution, and sensitivity, in the analysis of large mixtures of proteins. 2D-PAGE separates proteins by the sequential use at right angles of two different electrophoretic techniques based on two different properties of the proteins, the separation of the proteins according to their isoelectric point in the first dimension and the electrophoretic separation of the proteins in the presence of sodium dodecyl sulphate (SDS) according to their molecular weights in the Second dimension .
The detection of proteins after 2D-PAGE utilises either antibody-detection techniques or chemical stains such as coomassie blue or silver. The proteins on the gel are detected as distinct spots which can be assigned X and Y co-ordinates according to their isoelectric point (pI) and molecular weights (Mr), respectively. These co-ordinates can be used to identify proteins quickly and to compare gels without further analysis. 2D-PAGE was first developed by OFarrell (1975) but has subsequently undergone several modifications designed to improve its resolution, reliability and sensitivity.

I really like this presentation. But the music was very distracting. Voice, voice over, and music all were at different volumes. :(
