ok this is a fan made video. I love heroes and supernatural so I picked out my fave characters from each ad made this vid. You need to read this before watching the video for it to make sense.
Dean and Niki were in love but Dean found out she was cheating on him with DL. They are both in pain from the split and keep remembering the past. Dean mostly remembers her cheating on him but she remembers Dean. Dean decides to call Niki but he is unable to speak on the phone so Niki hangs up.
Niki then gets a video tape from Dean who has been kidnapped and runs to his rescue which result in her death as she is caught in the middle of an explosion. Dean is left heartbroken, remembering Niki as he leaves the city. Only at the end there is Niki, standing with a smile.
You can decide if that Niki is a spirit, or if Niki survived the blast. ;)
