Seagulls' sub Glenn Murray stepped off the bench to score two and make another goal to condemn Millwall to a second successive away defeat at a rain-swept Withdean Stadium.
Murray, a 28th-minute replacement for the injured Adam Virgo, fired the home side ahead a minute before the break against the run of play.
Tresor Kandol hauled The Lions level within three minutes of the restart only for Murray to add his second in highly contentious circumstances on 52 minutes.
Bradley Johnson (64mins) added a third before a Dean Cox cracker piled on the misery with Brighton's fourth nine minutes from time.
It had all started so brightly for The Lions with Dave Martin in particular, proving a real threat in the opening stages.
On two minutes the lively left winger intercepted a poor headed clearance, sprinted into the area and saw his cross cleared before firing a rasping drive over just 60 seconds later.
Lewis Grabban slipped a neat ball through to Chris Hackett after six minutes, with the latter's inviting cross inches away from the head of Tresor Kandol at the far post.
Millwall had a lucky let-off four minutes later when, with their first attack of the match, Brighton nearly broke the deadlock. Joe Anyinsah picked up the ball on the edge of the area and hit a fierce angled drive that smacked the foot of David Forde's left-hand post with the keeper beaten.
At the other end it was The Lions who continued to get into threatening positions, but lacked that incisive final touch where it mattered most.
Ali Fuseini, back in the team for suspended midfielder Jimmy Abdou, fired over from a Grabban corner before Grabban himself nutmegged Matt Richards and sent a low ball across the face of goal that was cleared by the boot of Andrew Whing.
But it was Brighton, who had offered little going forward throughout the opening half, who grabbed the lead against the run of play a minute before the break courtesy of Murray's clinical finish.
The Lions nearly levelled within 40 seconds of the second half when Neil Harris picked up a return pass from Frampton, created an opening and unleashed a right-foot effort that looked to be heading for the top corner. Unfortunately his effort clipped the woodwork and the danger was averted.
But just three minutes later The Lions were back on level terms. Harris slipped a neat pass through to Kandol, and the big striker despatched a low shot into the bottom right corner that gave John Sullivan no chance in the home goal.
Millwall were forced to make a change after 50 minutes when Marc Laird hobbled off with a hamstring injury and was replaced by Danny Spiller, making his first appearance of the season.
However within two minutes - and to the total bewilderment of the Millwall camp - Tony Craig was penalised for tugging Anyinsah's shirt when there was minimal, if any contact, between the two players as they jostled for the ball.
Referee Alan Wiley pointed to the spot and up stepped Murray to take the penalty. Although his spot-kick was saved low to keeper Forde's right, the sub reacted quickest and tucked the loose ball home to put Brighton 2-1 ahead.
Johnson increased their advantage just past the hour mark, capitalising on some poor defending before slotting home the Seagulls' third.
Even then, Millwall continued to press forward and a superb run and shot from Hackett could so easily have reduced the arrears 10 minutes from time, only for keeper Sullivan to deny the rampaging right-back.
But such was The Lions luck that every time they threatened to build up a head of steam back came Brighton, and that proved to be the case once again.
A quick break saw Murray burst down the right and send over an inch-perfect cross that was met first-time by the right boot of Cox who gave Forde no chance with a stunning first-time volley.
The diminutive little midfielder was denied a second goal four minutes from time, only for Forde to produce a magnificent tip-over from his 20-yard rocket, but by then the damage had already been done.
