They say "boy pee" as in he's still a child for not losing his virginity. The disease he was mentioning was a viral pimple that appears at the corner of the mouth when he gets too stressed (also mentions cracked corner), then show says if he got bit by a jellyfish, he'd need the urine too
then they mention how he'd be a faster learner of martial arts (younger people = more flexible) then they mention the puberty thing. Basically the big joke is just that he hasn't matured yet. ^^
