Hell, why not? Open C Major is fun as hell to use so I don't see why E Major wouldn't work.

Tuning to an E major chord by ear will result in a beautifully voiced chord, which properly emulates the naturally occurring harmonic series (R, 5, R, 3, 5, R). However, all other chords will sound out of tune, due to the fact that he's playing a fretted instrument, whose fret locations were determined using 12-tone equal temperament. So I doubt he actually can get good results playing exclusively in that tuning. Look up "just intonation" if you have further questions...
