Those are challenges that you can unlock on chapter 2; they reward goodies for completion.

I was wondering if the portal you walk into at 6:21 only works once? because i completed the game once.. Removed the savegame files and now im at this chapter again and the teleporter is gone?

If you have the Rod of trials from chapter 2, it should still work.

Where in chapter 2 do I find this Rod of trials?

On a shrine in front of the house where you find the claws. You have to kill all of the enemies first, then the shrine will lit up and the rod will appear.
