Didn't Work! Please help. All it did was get me stuck up the 1 block. And sometimes it would
just launch me through the wall and I would be stuck!. Or It would just put me to the regular boss room. Please Help!

Can people make their own sprites, enemies, weaknesses, weapons, bosses etc. in Construction Mode?

No, no, no, no, and no. In short: No. You're limited to about 90% of everything possible in the game. (Quite irritating that they didn't include electric pendulums in Timeman's tile set...)
