One of major lesions in DNA caused by alkylating mutagens is O6-methylguanine leading to mutations and cancerous cells. This is repaired by a ubiquitous suicidal enzyme, O6-methylguanine DNA methyltransferase (MGMT or AGT). This movie shows the structure of human enzyme bound and flip the methylguanine residue ready to accept the methyl group (1T38.pdb; Daniels et al. Nat.Struct.Mol.Biol. vol.11, 714).
