The engine has to be running at normal operating temperature so, no, you don't synchronize the carburetors with the enrichener on.
If you didn't change the synchronization or mixture screws and the engine won't run without the enrichener then synchronization is not the answer you're looking for. Synchronizing the carburetors is one of the last steps in tuning an XS1100. You have to find and fix any other engine problem or problems before you try to synchronize the carburetors.

All engines with multiple carburetors are similar, not just Japanese bikes.

First you adjust the synchronizer screws to set the carburetor throttle plates to the same throttle opening so they're all showing the same amount of vacuum, then you can set the idle mixture screws.
Start with the #3 carburetor.
#3 is the reference carburetor and is set with the master idle screw.
Next, adjust #4 to the same amount of vacuum as #3.
Move to the other side and adjust #1 to equal #2.
#2 carburetor screw sets both #1 and #2 to the #3 and #4 carburetor pair.

nice....you 1st moving pilot screws on each carbs...then you do somethings else...what you do moving the idle screw??
