Yes but both the matterand antimatter are being transformed so in this instance the absolute value of the antimatter would be used since positive and negative are nullified when transformed. So then, it would be 2(3g*300,000,000^2) Heating a gram of air by 3240000032 degrees. If my math is correct)

If you are referring to the video, the man on the horse represents antimatter.
If you are referring to E=MC2, it has everything to do with antimatter, because that is how matter is transformed into energy, which is what the theory is about

Perhaps but if I remember correctly my comment was directed at BillyTheSmartical who was apparently attempting to explain the video and was failing miserably at it. It is true that relativity and antimatter are relate but that relation would be similar to the relation the ground has to an apple because the tree grows in it. Relativity could be said to have relations with many topics but not many of those would be direct.

isnt it 6 grams ? the 3 of the anti matter and the 3 of the matter together makes it 6 ?!

the antimatter doesn't have grams, because when it combines with the matter, it doesnt add to it, it destroys it. if anythingit would be negative 3 grams.
also, in my above comment i wrote that it would be 900,000,000 joules. thats wrong because i forgot to square the speed of light.
it would actually release:
270,000,000,000,000,000 joules, enough energy to raise a small apple 1,678,060,910,000,000 miles into the air.
