I'd have to say it's a toss-up between Toad, Wario Bowser and Waluigi, but it you're meaning Mario characters that don't appear in Mario Party games, I'd have to add Rawk Hawk and Fawful to the list. Those characters make me laugh and I love funny characters.

If you mean for playing as in Mario Party, I tend to pick Peach simply because I have the best luck with her (like in these videos). She's luck a lucky charm for me.
