This video provides step-by-step instructions on how to isolate mononuclear cells from whole blood using a density gradient medium such as Ficoll-Paque™ PLUS. Watch the procedure being performed, which includes techniques such as layering diluted blood over Ficoll-Paque™ PLUS and collecting desired cells from the plasma/Ficoll-Paque™ PLUS interface. Ficoll-Paque™ PLUS can be used either prior to downstream cell separations with EasySep® or in conjunction with RosetteSep®.

  • I realized I made 5L's of PBS using a 1L recipe, thus my cells were being lysed since my PBS wasn't isotonic. Thank you for your advice though.

    spin at 800g and it will work fine, it won't make a difference to cells. I spin at 800g (for 20 min, brake off) all the time and i have have done 1000's of seperations.

    400g doesn't work for me when i have tried it. Most labs i know use 700-800g.

  • I followed this exact procedure, and for some reason failed to obtain a gradient and separation. Do you have any ideas as to why this occurred?
