I realized I made 5L's of PBS using a 1L recipe, thus my cells were being lysed since my PBS wasn't isotonic. Thank you for your advice though.

spin at 800g and it will work fine, it won't make a difference to cells. I spin at 800g (for 20 min, brake off) all the time and i have have done 1000's of seperations.
400g doesn't work for me when i have tried it. Most labs i know use 700-800g.

I followed this exact procedure, and for some reason failed to obtain a gradient and separation. Do you have any ideas as to why this occurred?
