The Caped Wonder takes on the Webslinger from New York in iconic Times Square.
Spider-Man moves around, a LOT, so fighting him can be very disorienting.
He's also very aggressive, and his webthrow will consistently catch you in the air. Superman's arctic breath, a move I rarely use, came in handy for this fight.

Don't forget this is a videogame. Their powers have been equalized. Superman still has more, but if the two have equal strength and speed, Spiderman would own this fight! Superman's vision and breathing powers made the difference! They even took away his flight, healing, and invulnerability! No way Superman could win under such circumstances! Ultimately, this isn't the comics (though it's fun to debate those).

All right. Good answer (for Spidey), assuming they're in the DCU. Kryptonite doesn't work every time, but it would even the odds enough for Superman to be beaten.
