Good to see that I'm not the only one who noticed that Jayne had unrequited feelings for Kaylee. I think that's why he was so antagonistic towards Simon: jealousy.

Jayne crushed on Kaylee. Kaylee crushed on Simon. Therefore Jayne wanted to crush Simon.
