DNA replication during S phase should occur only once for each segment of eukaryotic chromosome. Cdt 1 protein is a key component for the assembly of prereplicative complexes. The structure of Cdt 1 and two Geminin molecules can exist in two distinct forms, a permissive heterotrimer and an inhibitory heterohexamer, in which specific Cdt 1 residues important for the licensing are buried in the complex (2WVR.pdb: ref. De Marco et al. PNAS v. 106, 19807).
