Not really. Heath Ledgers Joker never laughed much, while Mark Hamills laughs at the end of almost every joke he says, stupid or not. Mark Hamill was my childhood voice for Joker, so bringing him back in the Arkham games was the best thing they could've done for someone to voice him in them. His voice matches the personality in this game amazingly.
