This is a time lapse of me working in my lab. For those in the know, I am doing a miniprep to extract bacterial plasmids from E. coli that contain full length HIV-1.

After I do the miniprep, I set up a digest on the DNA.

  • For a miniprep of only a the Env gene of HIV in a BL-2? I dont think so...

    For live virus in the BL-3? Absolutely.

  • are you using the QIAGEN kit? the box does look like it though :) ... and why aren't you wearing labcoat or glasses? I suppose an HIV lab should be at least a BL2 lab, right?

  • Its just ecoli with a gene from HIV in it. Because its noninfectious, it is a BL2.

    When working with infectious virus however, I do put a gown on and double glove and work in a BL3 lab.

  • Must not be that great of a lab if there are no mandatory regulations and a lab safety manager to enforce them..

  • Maybe you should following proper laboratory procedures and wear a lab coat.

  • but i think it's still good to always wear the lab coat because you never know what gets trapped on your clothes which you will be wearing for the rest of the day... especially if your lab isn't well ventilated

  • we also dont use glasses

  • ... buffer P2 has sodium hydroxide in it, buffer 3 has chaotrophic salts so glasses at least are a good idea. No matter, it's a fabulous video - just look after yourself!! :)
