This is where AE will warn you against adding vertexes. This is place where there is a difference between the skilled Rotoscope Artist and the Beginner. I run into this many instance a lot, You can spot my mistakes in the video. You have to take some serious time to analyze your video before taking a dive. If you run into this problem of adding multiple vertexes to one frame and quick changing shapes like Amoeba, End that mask at that frame and continue with a new one.

Go into your composition windows and turn on those keyframes. Click them all to be certain. Make sure you are working within the right Comp. Go to frame 5 and move the entire shape into it's new place. Wisely rotate to taste and move atleast 2 vertexes if not 3 to adjust for new shape. By the time you are done, The shape at Frame 5 should look just right. Now move on to frame 10 and do the same. You may run into a problem where the shape changes dramatically or requires more vertexes to be added

Of there is always a background in the mix. I just kept a reference background layer until I was ready to work on it. Before you start going into detail work, it is recommended that you finish your overall animation and make sure it works. Detail work are easy and fun, but they too can be time consuming.
So it is time for Keyframes and vertex fun. Remember, your goals and stick with them. Every 3-5 frames are guestimates I use, but there times during the time line where things will jump.
For the color, Coloring the rest of the body is straight foward at this point. Just color and leave detail. Ofcourse, if this is a production house, there would be strict adherence to color. I don't work in those enviroments. In regards to skin and makeup, the trick to this in AE is duplicating each layer and setting different opacity to archive the perfect color tone overall.

Now for the face. This is where you follow the same routine for each part of the body from frame 1 by outlining a facial feature. e.g eye, pupil, brow, lips, teeth, hair, eyelashes, blushes e.t.c You can go on if you like. Mask opacity and feather will come in really handy in animating these areas.
I think that pretty sums it for the rotoscoping part.

During extreme fast movement, the movie camera will capture motion blur. Like a fast hand movement will turn into fuzz. This is where you must use or animate mask feather and/or mask expansion. Always draw your vertexes along the inner lines of the fuzz shape instead of the outside when preparing for this. Regardless, I like to add a tiny bit of mask feather and expansion on all my shapes. You can also duplicate the fuzz effect using AE built motion blur and shutter. I never tried this.

Continue to move along the time line along 5 frames. You may sometimes hit a break where action freezes or slows down. You can increase the pace there by jumping more frames. Anyway, you're done with the main animation. Play it. If it lags in certain areas, you can force keyframes in the 'inbetween' keyrames and adjust shape to pleasure. I would recommend that you move a lot of vertexes at the same time if possible. You can also change the mask feather and expansion to taste in certain area.
