My first experience with the inoculation loop and bacterial cultures... I am seen here preparing a agar slant of Kleibsella pneumoniae from a pure culture.
@ledzep76 he didn't wear any glooves coz he is working in leminar flow cabinat which is microb free...and i think that he shoud have sterlize his hands by washing with ethanol......
1. after heating the loop u should also sterilize the agar plate. (even for just a second)
2. you should not open the plate in full because most common bacteria are aerobic therefore they can contaminate your culture. open the plate just enough for the inoculating loop.
3. after using the agar plate, place the plated in upward position (lid down technique) this is to prevent moisture to contaminate your culture.
No a lot of tube flaming going on. Fair enough he is working at a laminar flow cabinet but still your arms should be covered to protect you from any material drifting outwards. Unplugging of the tube is not the method I am used to: I would have the plug extracted by, and held by the same hand as the loop.

I agree with the other commenter. But I realize this was your first time.
It's also a good idea to not wave your loop around, as that increases your chances of collecting contaminants in the air.
