This is my Master's Research Project that was completed for my graduate degree (M.Sc.BMC) at the University of Toronto.

Description:
This animation describes one particular hypothesis regarding the development of opioid tolerance. The hypothesis supports the idea that a shift in equilibrium between the recycling and degradation of the opioid receptors may result in receptor down-regulation, which consequently results in tolerance.

I used Autodesk Maya 2009 and Adobe After Effects CS4 to create this project.

  • @brettis26 1) Abstinence (detox) or tapering off (alternate opioid maintenance, something natural and widely legal/available, as Kratom, or government controlled, as suboxone) 2) undiscovered epigenetic processes 3) rapid inverse signaling by co-administration of drug such as naloxone (non-detoxifying induced withdrawal) 4) NMDA antagonist drugs have been shown to lower tolerance (PCP, special K), 5) opioid peptides such as DAMGO seem to reverse sensitization (such as like an epigenetic process)

  • Thank you for this video. I'm using it to explain the similar process of benzodiazepine tolerance/GABA receptor down regulation to some folks.

  • Finally an answer, though in our modern technical age I cannot believe we still do not have any answer for such a relatively simple problem to study. Its not like its cancer with an unpredictable occurance. You can in fact enable this effect on cue, shows our priorities. Were much more interested in developing hydrogen bombs and ICBMs than medical research.

  • So emotional! =) Although better voice recording and not such a cruel voice would be nice, overall the vedeo is very emotional, and because of it - easy to remember. ^.^
