Blebbistatin is a small molecule inhibitor of non-muscle myosin II, which plays a critical role in cell migration, cell division, and many other cellular processes. The sudden inactivation of myosin II caused the extension of peripheral membrane in normal epithelial cells.
I'm guessing since blebbistatin inhibits cell contractility via myosin II pathway, the cells relax more and expand onto the substrate.
