It doesn't matter what connection do you have. I you have both - your phone OS will ask you which you wonna use.
Skype can perfectly work with GPRS/EDGE/CSD connections.

Skype uses J2ME/SIS to connect to the internet. It doesn't matter if its GPRS, or if its WiFi. It's just like other J2ME/SIS programs.
If your mobile OS provides menu for choosing which internet access Skype should use, you'll get that menu after starting and loging into Skype...
