Hi I noticed that punctuation marks go under the third line like a 'y' or a 'p'. Usally in writing it stays above the third line is there a real reason for doing this or is it just to be unique?

As other letters has a special place for themselves, the place of the punctuation marks is where you see in the clip.

does Copperplate need to be written holding the pen i that reverse angle? Or is that just prefrerence?

Hello, thank you very much for your message.
Yes it is necessary to hold the nib parallel with the slant lines but there is a kind of holders which is called oblique holder so it is not necessary for calligrapher to hold his or her holder in prallel form.
