To get this parcel, you have to go where the statue that only Thugee characters can activate.

Needed characters:
Thugee Character

Activate the shrine, and then fall down where the spikes used to be, go to the left, blow up the glass, take the parcel to the post box, save and exit and parcel collected!

If you mean other parcel's being enabled, yeah. If you use a code/cheat to unlock a parcel, no.
