Real time pcr is based on the principle of pcr, the difference is that in real time pcr we adding a dna probe which consists a short DNA complementary to target DNA  with one end is fluorescent reporter and another end is a quencher. now when the primer bind to target DNA as the chain is elongated until meet DNA probe, the 5'-3'-exonuclease will degrades the probe and separate reporter and quencher.
