Just like you said, the game comes out in two days; you cannot play Spore until then. If you preloaded the game, that just saves you time on release day that would otherwise have been spent downloading it.

It's only around 15 mins long, but there's actually been a lot of good things said about it. It's a good idea to go through each stage too, as they can give you unique items or bonuses that you wouldn't get if you skipped it.

Spore is playable forever once you reach the space stage; it plays out as a sandbox game (like GTA or Mercenaries 2)from there. Before that though, the cell stage is about 15 mins, and creature-civilization stages roughly are an hour longer than the last stage. The first time you play Spore, you have to play each stage at least 15 minutes before you can skip it. After you get to the space stage, you can go back to any of the other stages and play them again and for as long as you want to.
