  • @bored4longtimes yes, holes are punched to allow the antibody to enter the cell. That's why one of the cons of this technique is that the sample needs to be dead. This was what my lecturer told me :)

  • Just so ur aware of what you have done missy...i need to tell u this...is that this video has helped me a lot! I was studying microbio and I didn't understand the difference between normal flourescent microscopy and immunofluorescence...ty very much for this vid...ur the best!!

    You use something to 'permeabilize' the cell, like Triton X-100

  • I'm confused... Antibodies are huge (~150kDa) how do they get past the plasma membrane to label sub-cellular components?

    Are holes poked in the membrane during fixing?

  • Extremely helpful! I did immunofluorescence microscopy on microtubules in a lab and its really hard to tell where the division of the cells are :S
