Of course ;-) You just plug your keyboard midi output to the soft midi input (Zynaddsubfx or others) and the audio output of this software to the input of your audio card and the enjoy.
Try the soft "Patchage". This GUI can help you a lot to connect all your devices and softsynths more easily than Jack.
