While writing my first post I still had the anime ending in mind, only then I remembered that he's on a hill in the manga. My bad there.
Anyway, yes, her lifespan does matter. If she's destined to live for a thousand years, she WILL live for a thousand years. The only way to kill somebody before their lifespan runs out is by a Death Note.
