The overall goal of this procedure is to adapt human induced Pluripotent Stem (iPS) Cells to feeder-free culture using complete KnockOut™ Serum Replacement Feeder-Free medium (KSR-FF). This is accomplished by first preparing Geltrex-coated or CELLstart-coated plates, passaging your iPS from MEFs to feeder-free medium using dispase, and continually maintaining your iPS cells feeder-free. Ultimately, results can be obtained that show similar cell growth and stemness found with MEF cultures through immunostaining with pluripotent markers.
