How do you know which player has an advantage over the other?

Be able to recognized the matchups or mismatches. Differences in height, speed, quickness, low post offense, etc all play a part in which player has the advantage. Recognizing things like that one the fly must be done very quickly and are a big help.

I have one question is it better to have run the play set on manual or automatic?

Neither if you don't know how to run plays. If you don't know what the Cpu is running then don't put it on automatic.

how do you exploit the musmatch when a bigman is on your point gaurd like in this vid when tyrus thomas was on chauncey

Use IsoMotion moves and get by the slower defender. Also, if a big man is on your PG, then their PG is on one of your big men. This is the time to enter the ball into the low post and expose the mismatch another way.
