@colleenk127 When things are cold, it helps even more to keep things from dissolving in the liquid. Think about how long it takes to dissolve sugar in cold water vs. hot water.

Can yea still use an inoculating loop to remove them dna?

@sundaypiglet Yes - you can use a lot of different things to try to fish out the DNA. A wooden stick (like a BBQ skewer) is often useful because the DNA will stick to the wood pretty well.
