The briefcase contents are all things to all people, it is what you singularly desire the most which gives the purpose and reason for your existence, similairties are the ark of the covenant, spear of destiny and holy grail, material objects that we have lost that serve the purpose to making us whole again. The whole structure of the film is based around five ways of viewing the world and the quest that this entails, Authority, Honor, Mystery, Religion and Spirituality, Logic and Reason.

I don't believe for a second it's Marcell's soul. You can state the 666 combo for the briefcase, but when Mrs. Wallace and Vega return from their "date" she punches in the code for the sercuity, it kind of looks like she hits 666 in the code.
