This is a chromatography column that I did in Organic Chemistry lab. You can watch β-carotene and lycopene(to a lesser extent) travel through silica gel to be collected at the bottom.

Two solvents were used: hexane and a 9:1 mixture of hexane and ethyl acetate. The hexane drives the β-carotene down the column, then the 9:1 was added. Due to the polar nature of the 9:1 and lycopene, the red band begins to move more quickly upon the 9:1's addition. This can be observed in the video.

An hour of work for 40 seconds of video? It was worth it.

    Hmm, perhaps you're correct about the polarity. I'd check if

    1. organic wasn't done for the semester

    2. it wasn't 5:30am with me still having not been to bed; my chemistry skills fade late at night:)

  • What a shitty column run you did.

    The eluent is clearly not perpendicular to the column. You either did a shitty job packing the column or loading the column.

    Next time, cap the column and pump air into the column so you won't have to speed the run by 128x. You also get less band broadening.

  • dude, i think u meant lycopene is less polar. less polar travels faster in a silica gel column. unless if u'r using reverse phase silica gel.
