This is the opening scene of the first episode of the second season of Lost, "Man of Science, Man of Faith." It is one of my personal favorite shots of the entire show. This is the first time we see Desmond and what is inside the hatch.

Nobody noticed that at 3:19, the hatch hole is much bigger than the door we all know ?

As individual sequences go this was one of my favourites.
Along with:
1- Ending of season 1 where Jack and Locke are looking down the hatch.
2- Start of season 3 when we see the plane crash.
3- The end of Tabula Rasa, the beautiful Wash Away is playing and then the camera dollies around Locke sitting on the beach and the music goes all sinister.
Actually, damn there are too many. Great show.
